Diversidad genética de especies de Rubus determinada mediante RAPD

Resumen

La mora de castilla (Rubus glaucus Benth) pertenece a la familia de las Rosáceas y comprende numerosas especies cultivadas y silvestres.  En Venezuela se cultiva la especie R. glaucus en los estados Táchira, Mérida, Trujillo, Lara y Aragua. Algunas especies silvestres promisorias crecen cercanas a R. glaucus, entre ellas R. idaeus conocida como frambuesa. La falta de variedades comerciales de mora, así como la preservación de especies silvestres, plantea la necesidad de realizar  mejoramiento  genético.  El  objetivo  de este trabajo fue la amplificación del ADN mediante marcadores RAPD en especies del género Rubus de los estados Táchira y Aragua, para determinar la diversidad genética. El ADN se extrajo de hojas frescas de R. glaucus, R. idaeus y Rubus spp. Se utilizaron 23 iniciadores RAPD y se seleccionaron OPA10,  OPA18,  OPW06,  OPB09,  OPM10, OPK20, que produjeron 44 bandas de las cuales 38 fueron polimórficas. El iniciador OPW06 resultó el más informativo con un PIC de 0,38. Los análisis de agrupamiento y coordenadas principales coincidieron, discriminando la especie R. idaeus del resto de las especies, como también separaron los genotipos del estado Aragua de los del estado Táchira. La similitud entre genotipos en un mismo estado, sugiere la probable influencia del cruce de polen entre especies silvestres y cultivadas, así como el intercambio de material de propagación clonal utilizado por los productores. Estos resultados podrán ser utilizados en programas de conservación de especies y mejoramiento genético que incluyan cruzamientos interespecíficos
