RAPD analysis between and within populations derived from wild x cultivated sunflower

Abstract

This study aimed to estimate genetic relationships (RG) among 10 populations generated by crossing wild sunflower with HA-89 cultivated line, 26 RAPD markers were used. Plants were grown under greenhouse conditions to obtain young and healthy leaves, from which DNA was isolated following the protocol of Doyle and Doyle (1990). Isolated DNA was quantified and diluted to a concentration of 15 ng.ul-1, and then amplified in the presence of Taq polymerase with 30 random decameric primers from Operon Technologies. Amplification products were electrophoretically separated in 1,2% agarose gels. They were visualized and analyzed on a Biorad’s Chemidoc digitalyzer using the Quantity One ® software v. 4.2. Binary matrices (0,1) were made, from which statistical analysis was done. To estimate the genetic relationships within and between populations, a simple matching association coefficient was used, and phenetic relationships were represented in a dendrogram constructed with the Ward method. The population with H. deserticola pedigree was the one located closest to the cultivated line HA-89 and population with H. anomalus pedigree was the most distant.
